A native of Iowa, Brandon was born on October 9, 1979 in Des Moines and grew up in the city's suburb of Norwalk where he later attended Norwalk Senior High School while enthusiastically developed his acting skills at the Norwalk Theater of Performing Arts. Upon his graduation in 1997, he then enrolled in University of Iowa to pursue a degree in English, but as his desire to act became really hard to resist, the brown-eyed teen ultimately decided to give up his study after a year and so headed for Hollywood to search for good opportunity to flourish a career in entertainment industry.

Anxious to begin his career as a professional actor, Routh dropped out of his studies at the University of Iowa in 1999 to move to Los Angeles. His first role was a guest appearance on the ABC-TV sitcom Odd Man Out. In 2000, he became a regular on the MTV series Undressed. After a guest appearance on the hit series Gilmore Girls, Routh was chosen in May 2001 to originate the role of Seth Anderson on the daytime soap opera One Life to Live.

Brandon Routh was a young actor with only a few television credits when he was cast as Superman for the big-budget 2006 feature Superman Returns. Routh grew up in Iowa and attended the University of Iowa before leaving to make it as an actor. In Los Angeles he found work in television, and between 1999 and 2002 he appeared in Odd Man Out, Gilmore Girls and MTV's Undressed, and was a regular cast member of the daytime soap opera One Life to Live. Routh's only other feature film credit was the 2005 thriller Karla (sometimes titled Deadly), but Superman Returns director Bryan Singer pushed for an "unknown" actor and Routh won the role as the Man of Steel. Since then he's made several guest appearances on TV and appeared in the films Zack and Miri Make a Porno (2008, starring Seth Rogen) and Life is Hot in Cracktown (2009).
